種子擴大培養的任務
工gong業ye生sheng產chan規gui模mo越yue大da,每mei次ci發fa酵jiao所suo需xu的de種zhong子zi就jiu越yue多duo。要yao使shi小xiao小xiao的de微wei生sheng物wu在zai幾ji十shi小xiao時shi的de較jiao短duan時shi間jian內nei,完wan成cheng如ru此ci巨ju大da的de發fa酵jiao轉zhuan化hua任ren務wu,那na就jiu必bi須xu具ju備bei數shu量liang巨ju大da的de微wei生sheng物wu細xi胞bao才cai行xing。菌jun種zhong擴kuo大da培pei養yang的de目mu的de就jiu是shi要yao為wei每mei次ci發fa酵jiao罐guan的de投tou料liao提ti供gong相xiang當dang數shu量liang的de代dai謝xie旺wang盛sheng的de種zhong子zi。因yin為wei發fa酵jiao時shi間jian的de長chang短duan和he接jie種zhong量liang的de大da小xiao有you關guan,接jie種zhong量liang大da,發fa酵jiao時shi間jian則ze短duan。將jiang較jiao多duo數shu量liang的de成cheng熟shu菌jun體ti接jie入ru發fa酵jiao罐guan中zhong,就jiu有you利li於yu縮suo短duan發fa酵jiao時shi間jian,提ti高gao發fa酵jiao罐guan的de利li用yong率lv,並bing且qie也ye有you利li於yu減jian少shao染ran菌jun的de機ji會hui。因yin此ci,種zhong子zi擴kuo大da培pei養yang的de任ren務wu,不bu但dan要yao得de到dao純chun而er壯zhuang的de菌jun體ti,而er且qie還hai要yao獲huo得de活huo力li旺wang盛sheng的de、接jie種zhong數shu量liang足zu夠gou的de菌jun體ti。對dui於yu不bu同tong產chan品pin的de發fa酵jiao過guo程cheng來lai說shuo,必bi須xu根gen據ju菌jun種zhong生sheng長chang繁fan殖zhi速su度du快kuai慢man決jue定ding種zhong子zi擴kuo大da培pei養yang的de級ji數shu,抗kang生sheng素su生sheng產chan中zhong,放fang線xian菌jun的de細xi胞bao生sheng長chang繁fan殖zhi較jiao慢man,常chang常chang采cai用yong三san級ji種zhong子zi擴kuo大da培pei養yang。一yi般ban50 tfajiaoguanduocaiyongsanjifajiao,youdeshenzhicaiyongsijifajiao,rulianmeisushengchan。youxiemeizhijifajiaoshengchanyecaiyongsanjifajiao。erguansuanjiqitaanjisuandefajiaosuocaiyongdejunzhongshixijun,shengchangfanzhisuduhenkuai,yibancaiyongerjifajiao。
種子製備的過程
細菌、酵母菌的種子製備就是一個細胞數量增加的過程。細菌的斜麵培養基多采用碳源限量而氮源豐富的配方,牛肉膏、蛋白腖常用作有機氮源。細菌培養溫度大多數為37 ℃,少數為28 ℃,細菌菌體培養時間一般1~2天,產芽孢的細菌則需培養5~10天。
黴菌、放線菌的種子製備一般包括兩個過程,即在固體培養基上生產大量孢子的孢子製備和在液體培養基中生產大量菌絲的種子製備過程。
⒈ 孢子製備
孢子製備是種子製備的開始,是發酵生產的一個重要環節。孢子的質量、數量對以後菌絲的生長、繁殖和發酵產量都有明顯的影響。不同菌種的孢子製備工藝有其不同的特點。
⑴ 放線菌孢子的製備 放線菌的孢子培養一般采用瓊脂斜麵培養基,培養基中含有一些適合產孢子的營養成分,如麩皮、豌豆浸汁、蛋白腖和一些無機鹽等。碳源和氮源不要太豐富(碳源約為1%,氮源不超過0.5%),碳(tan)源(yuan)豐(feng)富(fu)容(rong)易(yi)造(zao)成(cheng)生(sheng)理(li)酸(suan)性(xing)的(de)營(ying)養(yang)環(huan)境(jing),不(bu)利(li)於(yu)放(fang)線(xian)菌(jun)孢(bao)子(zi)的(de)形(xing)成(cheng),氮(dan)源(yuan)豐(feng)富(fu)則(ze)有(you)利(li)於(yu)菌(jun)絲(si)繁(fan)殖(zhi)而(er)不(bu)利(li)於(yu)孢(bao)子(zi)形(xing)成(cheng)。一(yi)般(ban)情(qing)況(kuang)下(xia),幹(gan)燥(zao)和(he)限(xian)製(zhi)營(ying)養(yang)可(ke)直(zhi)接(jie)或(huo)間(jian)接(jie)誘(you)導(dao)孢(bao)子(zi)形(xing)成(cheng)。放(fang)線(xian)菌(jun)斜(xie)麵(mian)的(de)培(pei)養(yang)溫(wen)度(du)大(da)多(duo)數(shu)為(wei)28 ℃,少數為37 ℃,培養時間為5~14天。

⑵ 黴菌孢子的製備 黴菌的孢子培養,一般以大米、小米、玉米、麩皮、麥mai粒li等deng天tian然ran農nong產chan品pin為wei培pei養yang基ji。這zhe是shi由you於yu這zhe些xie農nong產chan品pin中zhong的de營ying養yang成cheng分fen較jiao適shi合he黴mei菌jun的de孢bao子zi繁fan殖zhi,而er且qie這zhe類lei培pei養yang基ji的de表biao麵mian積ji較jiao大da,可ke獲huo得de大da量liang的de孢bao子zi。黴mei菌jun的de培pei養yang一yi般ban為wei25~28 ℃,培養時間為4~14天。
⒉ 種子製備
種(zhong)子(zi)製(zhi)備(bei)是(shi)將(jiang)固(gu)體(ti)培(pei)養(yang)基(ji)上(shang)培(pei)養(yang)出(chu)的(de)孢(bao)子(zi)或(huo)菌(jun)體(ti)轉(zhuan)入(ru)到(dao)液(ye)體(ti)培(pei)養(yang)基(ji)中(zhong)培(pei)養(yang),使(shi)其(qi)繁(fan)殖(zhi)成(cheng)大(da)量(liang)菌(jun)絲(si)或(huo)菌(jun)體(ti)的(de)過(guo)程(cheng)。種(zhong)子(zi)製(zhi)備(bei)所(suo)使(shi)用(yong)的(de)培(pei)養(yang)基(ji)和(he)其(qi)他(ta)工(gong)藝(yi)條(tiao)件(jian),都(dou)要(yao)有(you)利(li)於(yu)孢(bao)子(zi)發(fa)芽(ya)、菌絲繁殖或菌體增殖。
⑴ 搖瓶種子製備 mouxiebaozifayahejunsifanzhisuduhuanmandejunzhong,xujiangbaozijingyaopingpeiyangchengjunsihouzaijinruzhongziguan,zhejiushiyaopingzhongzi。yaopingxiangdangyuweisuoledezhongziguan,qipeiyangjipeifanghepeiyangtiaojianyuzhongziguanxiangsi。
搖瓶種子進罐,常采用母瓶、zipingliangjipeiyang,youshimupingzhongziyekeyizhijiejinguan。zhongzipeiyangjiyaoqiubijiaofengfuhewanquan,bingyibeijuntifenjieliyong,danyuanfengfuyouliyujunsishengchang。yuanzeshanggezhongyingyangchengfenbuyiguonong,zipingpeiyangjinongdubimupinglvegao,gengjiejinzhongziguandepeiyangjipeifang。
⑵ 種子罐種子製備 種子罐種子製備的工藝過程,因菌種不同而異,一般可分為一級種子、二級種子和三級種子的製備。孢子(或搖瓶菌絲)被(bei)接(jie)入(ru)到(dao)體(ti)積(ji)較(jiao)小(xiao)的(de)種(zhong)子(zi)罐(guan)中(zhong),經(jing)培(pei)養(yang)後(hou)形(xing)成(cheng)大(da)量(liang)的(de)菌(jun)絲(si),這(zhe)樣(yang)的(de)種(zhong)子(zi)稱(cheng)為(wei)一(yi)級(ji)種(zhong)子(zi),把(ba)一(yi)級(ji)種(zhong)子(zi)轉(zhuan)入(ru)發(fa)酵(jiao)罐(guan)內(nei)發(fa)酵(jiao),稱(cheng)為(wei)二(er)級(ji)發(fa)酵(jiao)。如(ru)果(guo)將(jiang)一(yi)級(ji)種(zhong)子(zi)接(jie)入(ru)體(ti)積(ji)較(jiao)大(da)的(de)種(zhong)子(zi)罐(guan)內(nei),經(jing)過(guo)培(pei)養(yang)形(xing)成(cheng)更(geng)多(duo)的(de)菌(jun)絲(si),這(zhe)樣(yang)製(zhi)備(bei)的(de)種(zhong)子(zi)稱(cheng)為(wei)二(er)級(ji)種(zhong)子(zi),將(jiang)二(er)級(ji)種(zhong)子(zi)轉(zhuan)入(ru)發(fa)酵(jiao)罐(guan)內(nei)發(fa)酵(jiao),稱(cheng)為(wei)三(san)級(ji)發(fa)酵(jiao)。同(tong)樣(yang)道(dao)理(li),使(shi)用(yong)三(san)級(ji)種(zhong)子(zi)的(de)發(fa)酵(jiao),稱(cheng)為(wei)四(si)級(ji)發(fa)酵(jiao)。
種zhong子zi罐guan的de級ji數shu主zhu要yao決jue定ding於yu菌jun種zhong的de性xing質zhi和he菌jun體ti生sheng長chang速su度du及ji發fa酵jiao設she備bei的de合he理li應ying用yong。種zhong子zi製zhi備bei的de目mu的de是shi要yao形xing成cheng一yi定ding數shu量liang和he質zhi量liang的de菌jun體ti。孢bao子zi發fa芽ya和he菌jun體ti開kai始shi繁fan殖zhi時shi,菌jun體ti量liang很hen少shao,在zai小xiao型xing罐guan內nei即ji可ke進jin行xing。發fa酵jiao的de目mu的de是shi獲huo得de大da量liang的de發fa酵jiao產chan物wu。產chan物wu是shi在zai菌jun體ti大da量liang形xing成cheng並bing達da到dao一yi定ding生sheng長chang階jie段duan後hou形xing成cheng的de,需xu要yao在zai大da型xing發fa酵jiao罐guan內nei才cai能neng進jin行xing。同tong時shi若ruo幹gan發fa酵jiao產chan物wu的de產chan生sheng菌jun,其qi不bu同tong生sheng長chang階jie段duan對dui營ying養yang和he培pei養yang條tiao件jian的de要yao求qiu有you差cha異yi。因yin此ci,將jiang兩liang個ge目mu的de不bu同tong、工gong藝yi要yao求qiu有you差cha異yi的de生sheng物wu學xue過guo程cheng放fang在zai一yi個ge大da罐guan內nei進jin行xing,既ji影ying響xiang發fa酵jiao產chan物wu的de產chan量liang,又you會hui造zao成cheng動dong力li和he設she備bei的de浪lang費fei。種zhong子zi罐guan級ji數shu減jian少shao,有you利li於yu生sheng產chan過guo程cheng的de簡jian化hua及ji發fa酵jiao過guo程cheng的de控kong製zhi,可ke以yi減jian少shao因yin種zhong子zi生sheng長chang異yi常chang而er造zao成cheng發fa酵jiao的de波bo動dong。
種子培養

液(ye)體(ti)深(shen)層(ceng)種(zhong)子(zi)罐(guan)從(cong)罐(guan)底(di)部(bu)通(tong)氣(qi),送(song)入(ru)的(de)空(kong)氣(qi)由(you)攪(jiao)拌(ban)槳(jiang)葉(ye)分(fen)散(san)成(cheng)微(wei)小(xiao)氣(qi)泡(pao)以(yi)促(cu)進(jin)氧(yang)的(de)溶(rong)解(jie)。這(zhe)種(zhong)由(you)罐(guan)底(di)部(bu)通(tong)氣(qi)攪(jiao)拌(ban)的(de)培(pei)養(yang)方(fang)法(fa),相(xiang)對(dui)於(yu)由(you)氣(qi)液(ye)界(jie)麵(mian)靠(kao)自(zi)然(ran)擴(kuo)散(san)使(shi)氧(yang)溶(rong)解(jie)的(de)表(biao)麵(mian)培(pei)養(yang)法(fa)來(lai)講(jiang),稱(cheng)為(wei)深(shen)層(ceng)培(pei)養(yang)法(fa)。其(qi)特(te)點(dian)是(shi)容(rong)易(yi)按(an)照(zhao)生(sheng)產(chan)菌(jun)種(zhong)對(dui)於(yu)代(dai)謝(xie)的(de)營(ying)養(yang)要(yao)求(qiu)以(yi)及(ji)不(bu)同(tong)生(sheng)理(li)時(shi)期(qi)的(de)通(tong)氣(qi)、攪拌、溫度與培養基中氫離子濃度等條件,選擇最佳培養條件。
⑴ 深層培養基本操作的三個控製點
① 滅菌 發(fa)酵(jiao)工(gong)業(ye)要(yao)求(qiu)純(chun)培(pei)養(yang),因(yin)此(ci)在(zai)種(zhong)子(zi)培(pei)養(yang)前(qian)必(bi)須(xu)對(dui)培(pei)養(yang)基(ji)進(jin)行(xing)加(jia)熱(re)滅(mie)菌(jun)。所(suo)以(yi)種(zhong)子(zi)罐(guan)具(ju)有(you)蒸(zheng)汽(qi)夾(jia)套(tao),以(yi)便(bian)將(jiang)培(pei)養(yang)基(ji)和(he)種(zhong)子(zi)罐(guan)進(jin)行(xing)加(jia)熱(re)滅(mie)菌(jun),或(huo)者(zhe)將(jiang)培(pei)養(yang)基(ji)由(you)連(lian)續(xu)加(jia)熱(re)滅(mie)菌(jun)器(qi)滅(mie)菌(jun),並(bing)連(lian)續(xu)地(di)輸(shu)送(song)於(yu)種(zhong)子(zi)罐(guan)內(nei)。
② 溫度控製 培(pei)養(yang)基(ji)滅(mie)菌(jun)後(hou),冷(leng)卻(que)至(zhi)培(pei)養(yang)溫(wen)度(du)進(jin)行(xing)種(zhong)子(zi)培(pei)養(yang),由(you)於(yu)隨(sui)著(zhe)微(wei)生(sheng)物(wu)的(de)生(sheng)長(chang)和(he)繁(fan)殖(zhi)會(hui)產(chan)生(sheng)熱(re)量(liang),攪(jiao)拌(ban)也(ye)會(hui)產(chan)生(sheng)熱(re)量(liang),所(suo)以(yi)要(yao)維(wei)持(chi)溫(wen)度(du)恒(heng)定(ding),需(xu)在(zai)夾(jia)套(tao)中(zhong)或(huo)盤(pan)管(guan)中(zhong)通(tong)冷(leng)卻(que)水(shui)循(xun)環(huan)。
③ 通氣、攪拌 空kong氣qi進jin入ru種zhong子zi罐guan前qian先xian經jing過guo空kong氣qi過guo濾lv器qi除chu去qu雜za菌jun,製zhi成cheng無wu菌jun空kong氣qi,而er後hou由you罐guan底di部bu進jin入ru,再zai通tong過guo攪jiao拌ban將jiang空kong氣qi分fen散san成cheng微wei小xiao氣qi泡pao。為wei了le延yan長chang氣qi泡pao滯zhi留liu時shi間jian,可ke在zai罐guan內nei裝zhuang擋dang板ban產chan生sheng渦wo流liu。攪jiao拌ban的de目mu的de除chu增zeng加jia溶rong解jie氧yang以yi外wai,可ke使shi培pei養yang液ye中zhong的de微wei生sheng物wu均jun勻yun地di分fen散san在zai種zhong子zi罐guan內nei,促cu進jin熱re傳chuan遞di,以yi及jipH均充,並使加入的酸和堿均勻分散等。
⑵ 幾種深層培養法
① 控製培養法 根(gen)據(ju)罐(guan)內(nei)部(bu)的(de)變(bian)化(hua)情(qing)況(kuang),掌(zhang)握(wo)短(duan)暫(zan)時(shi)間(jian)內(nei)狀(zhuang)態(tai)變(bian)量(liang)的(de)變(bian)化(hua)以(yi)及(ji)可(ke)能(neng)測(ce)定(ding)的(de)環(huan)境(jing)因(yin)子(zi)對(dui)微(wei)生(sheng)物(wu)代(dai)謝(xie)活(huo)動(dong)的(de)影(ying)響(xiang),並(bing)以(yi)此(ci)為(wei)基(ji)礎(chu)進(jin)行(xing)控(kong)製(zhi)培(pei)養(yang),以(yi)達(da)到(dao)產(chan)物(wu)的(de)最(zui)優(you)培(pei)養(yang)條(tiao)件(jian)。為(wei)此(ci),用(yong)測(ce)定(ding)狀(zhuang)態(tai)變(bian)量(liang)的(de)傳(chuan)感(gan)器(qi)取(qu)得(de)數(shu)據(ju),經(jing)電(dian)子(zi)計(ji)算(suan)機(ji)進(jin)行(xing)綜(zong)合(he)分(fen)析(xi),再(zai)將(jiang)其(qi)結(jie)果(guo)作(zuo)為(wei)反(fan)饋(kui)調(tiao)節(jie)的(de)信(xin)號(hao),將(jiang)環(huan)境(jing)(培養條件)控製於給定的基準內。這就叫做電子計算機控製培養。目前已大量用於露天大罐啤酒發酵。
② 載體培養法 載(zai)體(ti)培(pei)養(yang)法(fa)脫(tuo)胎(tai)於(yu)曲(qu)法(fa)培(pei)養(yang),同(tong)時(shi)又(you)吸(xi)收(shou)了(le)液(ye)體(ti)培(pei)養(yang)的(de)優(you)點(dian),是(shi)近(jin)年(nian)來(lai)新(xin)發(fa)展(zhan)的(de)一(yi)種(zhong)培(pei)養(yang)方(fang)法(fa)。特(te)征(zheng)是(shi)以(yi)天(tian)然(ran)或(huo)人(ren)工(gong)合(he)成(cheng)的(de)多(duo)孔(kong)材(cai)料(liao)代(dai)替(ti)麩(fu)皮(pi)之(zhi)類(lei)的(de)固(gu)體(ti)基(ji)質(zhi)作(zuo)為(wei)微(wei)生(sheng)物(wu)的(de)載(zai)體(ti),營(ying)養(yang)成(cheng)分(fen)可(ke)以(yi)嚴(yan)格(ge)控(kong)製(zhi)。發(fa)酵(jiao)結(jie)束(shu),隻(zhi)要(yao)將(jiang)菌(jun)體(ti)和(he)培(pei)養(yang)基(ji)擠(ji)壓(ya)出(chu)來(lai)進(jin)行(xing)抽(chou)提(ti),載(zai)體(ti)又(you)可(ke)以(yi)重(zhong)新(xin)使(shi)用(yong)。

種子質量的控製
種zhong子zi質zhi量liang是shi影ying響xiang發fa酵jiao生sheng產chan水shui平ping的de重zhong要yao因yin素su。種zhong子zi質zhi量liang的de優you劣lie,主zhu要yao取qu決jue於yu菌jun種zhong本ben身shen的de遺yi傳chuan特te性xing和he培pei養yang條tiao件jian兩liang個ge方fang麵mian。這zhe就jiu是shi說shuo既ji要yao有you優you良liang的de菌jun種zhong,又you要yao有you良liang好hao的de培pei養yang條tiao件jian才cai能neng獲huo得de高gao質zhi量liang的de種zhong子zi。
㈠ 影響孢子質量的因素及其控製
孢子質量與培養基、培養溫度、濕度、培養時間、接種量等有關,這些因素相互聯係、相互影響,因此必須全麵考慮各種因素,認真加以控製。
⒈ 培養基
構成孢子培養基的原材料,其產地、品種、加工方法和用量對孢子質量都有一定的影響。生產過程中孢子質量不穩定的現象,常常是原材料質量不穩定所造成的。原材料產地、品pin種zhong和he加jia工gong方fang法fa的de不bu同tong,會hui導dao致zhi培pei養yang基ji中zhong的de微wei量liang元yuan素su和he其qi他ta營ying養yang成cheng分fen含han量liang的de變bian化hua。例li如ru,由you於yu生sheng產chan蛋dan白bai腖dong所suo用yong的de原yuan材cai料liao及ji生sheng產chan工gong藝yi的de不bu同tong,蛋dan白bai腖dong的de微wei量liang元yuan素su含han量liang、磷含量、氨(an)基(ji)酸(suan)組(zu)分(fen)均(jun)有(you)所(suo)不(bu)同(tong),而(er)這(zhe)些(xie)營(ying)養(yang)成(cheng)分(fen)對(dui)於(yu)菌(jun)體(ti)生(sheng)長(chang)和(he)孢(bao)子(zi)形(xing)成(cheng)有(you)重(zhong)要(yao)作(zuo)用(yong)。瓊(qiong)脂(zhi)的(de)牌(pai)號(hao)不(bu)同(tong),對(dui)孢(bao)子(zi)質(zhi)量(liang)也(ye)有(you)影(ying)響(xiang),這(zhe)是(shi)由(you)於(yu)不(bu)同(tong)牌(pai)號(hao)的(de)瓊(qiong)脂(zhi)含(han)有(you)不(bu)同(tong)的(de)無(wu)機(ji)離(li)子(zi)造(zao)成(cheng)的(de)。
此外,水質的影響也不能忽視。地區的不同、季(ji)節(jie)的(de)變(bian)化(hua)和(he)水(shui)源(yuan)的(de)汙(wu)染(ran),均(jun)可(ke)成(cheng)為(wei)水(shui)質(zhi)波(bo)動(dong)的(de)原(yuan)因(yin)。為(wei)了(le)避(bi)免(mian)水(shui)質(zhi)波(bo)動(dong)對(dui)孢(bao)子(zi)質(zhi)量(liang)的(de)影(ying)響(xiang),可(ke)在(zai)蒸(zheng)餾(liu)水(shui)或(huo)無(wu)鹽(yan)水(shui)中(zhong)加(jia)入(ru)適(shi)量(liang)的(de)無(wu)機(ji)鹽(yan),供(gong)配(pei)製(zhi)培(pei)養(yang)基(ji)使(shi)用(yong)。例(li)如(ru)在(zai)配(pei)製(zhi)生(sheng)產(chan)四(si)環(huan)素(su)的(de)斜(xie)麵(mian)培(pei)養(yang)基(ji)時(shi),有(you)時(shi)在(zai)無(wu)鹽(yan)水(shui)內(nei)加(jia)入(ru)0.03%(NH4)2HPO4,0.028%KH2PO4及0.01%MgSO4,確保孢子質量,提高四環素發酵產量。
為了保證孢子培養基的質量,斜麵培養基所用的主要原材料,糖、氮、磷(lin)含(han)量(liang)需(xu)經(jing)過(guo)化(hua)學(xue)分(fen)析(xi)及(ji)搖(yao)瓶(ping)發(fa)酵(jiao)試(shi)驗(yan)合(he)格(ge)後(hou)才(cai)能(neng)使(shi)用(yong)。製(zhi)備(bei)培(pei)養(yang)基(ji)時(shi)要(yao)嚴(yan)格(ge)控(kong)製(zhi)滅(mie)菌(jun)後(hou)的(de)培(pei)養(yang)基(ji)質(zhi)量(liang)。斜(xie)麵(mian)培(pei)養(yang)基(ji)使(shi)用(yong)前(qian),需(xu)在(zai)適(shi)當(dang)溫(wen)度(du)下(xia)放(fang)置(zhi)一(yi)定(ding)的(de)時(shi)間(jian),使(shi)斜(xie)麵(mian)無(wu)冷(leng)凝(ning)水(shui)呈(cheng)現(xian),水(shui)分(fen)適(shi)中(zhong)有(you)利(li)於(yu)孢(bao)子(zi)生(sheng)長(chang)。
peizhibaozipeiyangjihaiyinggaikaolvbutongdaixieleixingdejunluoduiduozhonganjisuandexuanze。junzhongzaigutipeiyangjishangkechengxianduozhongbutongdaixieleixingdejunluo,gezhonganjisuanduijunluodebiaoxianbutong。danyuanpinzhongyueduo,chuxiandejunluoleixingyeyueduo,buliyushengchandewending。xiemianpeiyangjishangyongjiaodanyidedanyuan,keyizhimouxiebuzhengchangxingjunluodechuxian;而對分離篩選的平板培養基則需加入較複雜的氮源,使其多種菌落類型充分表現,以利篩選。因此在製備固體培養基時有兩條經驗:① 供生產用的孢子培養基或作為製備砂土孢子或傳代所用的培養基要用比較單一的氮源,以便保持正常菌落類型的優勢;② 作為選種或分離用的平板培養基,則需采用較複雜的有機氮源,目的是便於選擇特殊代謝的菌落。
⒉ 培養溫度和濕度
weishengwuzaiyigejiaokuandewendufanweineishengchang。danshi,yaohuodegaozhiliangdebaozi,qizuishiwenduqujianhenxiazhai。yibanlaishuo,tigaopeiyangwendu,keshijuntidaixiehuodongjiakuai,suoduanpeiyangshijian,danshi,juntidetangdaixiehedandaixiedegezhongmeilei,duiwendudeminganxingshibutongde。yinci,peiyangwendubutong,jundeshenglizhuangtaiyebutong,ruguobushiyongzuishiwendupeiyangdebaozi,qishengchannenglijiuhuixiajiang。butongdejunzhuyaoqiudezuishiwendubutong,xujingshijiankaochaqueding。liru,guilielianmeijunxiemianzuishiwenduwei36.5~37 ℃,如果高於37 ℃,則孢子成熟早,易老化,接入發酵罐後,就會出現菌絲對糖、氮利用緩慢,氨基氮回升提前,發酵產量降低等現象。培養溫度控製低一些,則有利於孢子的形成。龜裂鏈黴菌斜麵先放在36.5 ℃培養3天,再放在28.5 ℃培養1天,所得的孢子數量比在36.5 ℃培養4天所得的孢子數量增加3~7倍。
斜麵孢子培養時,培養室的相對濕度對孢子形成的速度、數量和質量有很大影響。空氣中相對濕度高時,培養基內的水分蒸發少;相(xiang)對(dui)濕(shi)度(du)低(di)時(shi),培(pei)養(yang)基(ji)內(nei)的(de)水(shui)分(fen)蒸(zheng)發(fa)多(duo)。例(li)如(ru),在(zai)我(wo)國(guo)北(bei)方(fang)幹(gan)燥(zao)地(di)區(qu),冬(dong)季(ji)由(you)於(yu)氣(qi)候(hou)幹(gan)燥(zao),空(kong)氣(qi)相(xiang)對(dui)濕(shi)度(du)偏(pian)低(di),斜(xie)麵(mian)培(pei)養(yang)基(ji)內(nei)的(de)水(shui)分(fen)蒸(zheng)發(fa)的(de)快(kuai),致(zhi)使(shi)斜(xie)麵(mian)下(xia)部(bu)含(han)有(you)一(yi)定(ding)水(shui)分(fen),而(er)上(shang)部(bu)易(yi)幹(gan)癟(bie),這(zhe)時(shi)孢(bao)子(zi)長(chang)得(de)快(kuai),且(qie)從(cong)斜(xie)麵(mian)下(xia)部(bu)向(xiang)上(shang)長(chang)。夏(xia)季(ji)時(shi)空(kong)氣(qi)相(xiang)對(dui)濕(shi)度(du)高(gao),斜(xie)麵(mian)內(nei)水(shui)分(fen)蒸(zheng)發(fa)得(de)慢(man),這(zhe)時(shi)斜(xie)麵(mian)孢(bao)子(zi)從(cong)上(shang)部(bu)往(wang)下(xia)長(chang),下(xia)部(bu)常(chang)因(yin)積(ji)存(cun)冷(leng)凝(ning)水(shui),致(zhi)使(shi)孢(bao)子(zi)生(sheng)長(chang)得(de)慢(man)或(huo)孢(bao)子(zi)不(bu)能(neng)生(sheng)長(chang)。試(shi)驗(yan)表(biao)明(ming),在(zai)一(yi)定(ding)條(tiao)件(jian)下(xia)培(pei)養(yang)斜(xie)麵(mian)孢(bao)子(zi)時(shi),在(zai)北(bei)方(fang)相(xiang)對(dui)濕(shi)度(du)控(kong)製(zhi)在(zai)40%~45%,而在南方相對濕度控製在35%~42%,所得孢子質量較好。一般來說,真菌對濕度要求偏高,而放線菌對濕度要求偏低。
在(zai)培(pei)養(yang)箱(xiang)培(pei)養(yang)時(shi),如(ru)果(guo)相(xiang)對(dui)濕(shi)度(du)偏(pian)低(di),可(ke)放(fang)入(ru)盛(sheng)水(shui)的(de)平(ping)皿(min),提(ti)高(gao)培(pei)養(yang)箱(xiang)內(nei)的(de)相(xiang)對(dui)濕(shi)度(du),為(wei)了(le)保(bao)證(zheng)新(xin)鮮(xian)空(kong)氣(qi)的(de)交(jiao)換(huan),培(pei)養(yang)箱(xiang)每(mei)天(tian)宜(yi)開(kai)啟(qi)幾(ji)次(ci),以(yi)利(li)於(yu)孢(bao)子(zi)生(sheng)長(chang)。現(xian)代(dai)化(hua)的(de)培(pei)養(yang)箱(xiang)是(shi)恒(heng)溫(wen)、恒濕,並可換氣,不用人工控製。
最適培養溫度和濕度是相對的,例如相對濕度、培養基組分不同,對微生物的最適溫度會有影響。培養溫度、培養基組分不同也會影響到微生物培養的最適相對濕度。
⒊ 培養時間和冷藏時間
絲狀菌在斜麵培養基上的生長發育過程可分為五個階段:① 孢子發芽和基內菌絲生長階段;② 氣生菌絲生長階段;③ 孢子形成階段;④ 孢子成熟階段;⑤ 斜麵衰老菌絲自溶階段。
⑴ 孢子的培養時間 基ji內nei菌jun絲si和he氣qi生sheng菌jun絲si內nei部bu的de核he物wu質zhi和he細xi胞bao質zhi處chu於yu流liu動dong狀zhuang態tai,如ru果guo把ba菌jun絲si斷duan開kai,各ge菌jun絲si片pian斷duan之zhi間jian的de內nei容rong是shi不bu同tong的de,有you的de片pian斷duan中zhong含han有you核he粒li,有you的de片pian斷duan中zhong沒mei有you核he粒li,而er核he粒li的de多duo少shao亦yi不bu均jun勻yun,該gai階jie段duan的de菌jun絲si不bu適shi宜yi於yu菌jun種zhong保bao存cun和he傳chuan代dai。而er孢bao子zi本ben身shen是shi一yi個ge獨du立li的de遺yi傳chuan體ti,其qi遺yi傳chuan物wu質zhi比bi較jiao完wan整zheng,因yin此ci孢bao子zi用yong於yu傳chuan代dai和he保bao存cun均jun能neng保bao持chi原yuan始shi菌jun種zhong的de基ji本ben特te征zheng。但dan是shi孢bao子zi本ben身shen亦yi有you年nian輕qing與yu衰shuai老lao的de區qu別bie。一yi般ban來lai說shuo衰shuai老lao的de孢bao子zi不bu如ru年nian輕qing孢bao子zi,因yin為wei衰shuai老lao的de孢bao子zi已yi在zai逐zhu步bu進jin入ru發fa芽ya階jie段duan,核he物wu質zhi趨qu於yu分fen化hua狀zhuang態tai。孢bao子zi的de培pei養yang工gong藝yi一yi般ban選xuan擇ze在zai孢bao子zi成cheng熟shu階jie段duan時shi終zhong止zhi培pei養yang,此ci時shi顯xian微wei鏡jing下xia可ke見jian到dao成cheng串chuan孢bao子zi或huo遊you離li的de分fen散san孢bao子zi,如ru果guo繼ji續xu培pei養yang,則ze進jin入ru斜xie麵mian衰shuai老lao菌jun絲si自zi溶rong階jie段duan,表biao現xian為wei斜xie麵mian外wai觀guan變bian色se、發暗或黃、菌層下陷、有(you)時(shi)出(chu)現(xian)白(bai)色(se)斑(ban)點(dian)或(huo)發(fa)黑(hei)。白(bai)斑(ban)表(biao)示(shi)孢(bao)子(zi)發(fa)芽(ya)長(chang)出(chu)第(di)二(er)代(dai)菌(jun)絲(si),黑(hei)色(se)顯(xian)示(shi)菌(jun)絲(si)自(zi)溶(rong)。孢(bao)子(zi)的(de)培(pei)養(yang)時(shi)間(jian)對(dui)孢(bao)子(zi)質(zhi)量(liang)有(you)重(zhong)要(yao)影(ying)響(xiang),過(guo)於(yu)年(nian)輕(qing)的(de)孢(bao)子(zi)經(jing)不(bu)起(qi)冷(leng)藏(zang),如(ru)土(tu)黴(mei)素(su)菌(jun)種(zhong)斜(xie)麵(mian)培(pei)養(yang)4.5天,孢子尚未完全成熟,冷藏7~8天菌絲即開始自溶。而培養時間延長半天(即培養5天),孢子完全成熟,可冷藏20天也不自溶。過於衰老的孢子會導致生產能力下降,孢子的培養時間應控製在孢子量多、孢子成熟、發酵產量正常的階段終止培養。
⑵ 孢子的冷藏時間 斜(xie)麵(mian)孢(bao)子(zi)的(de)冷(leng)藏(zang)時(shi)間(jian),對(dui)孢(bao)子(zi)質(zhi)量(liang)也(ye)有(you)影(ying)響(xiang),其(qi)影(ying)響(xiang)隨(sui)菌(jun)種(zhong)不(bu)同(tong)而(er)異(yi),總(zong)的(de)原(yuan)則(ze)是(shi)冷(leng)藏(zang)時(shi)間(jian)宜(yi)短(duan)不(bu)宜(yi)長(chang)。曾(zeng)有(you)報(bao)道(dao),在(zai)鏈(lian)黴(mei)素(su)生(sheng)產(chan)中(zhong),斜(xie)麵(mian)孢(bao)子(zi)在(zai)6 ℃冷藏2個月後的發酵單位比冷藏1個月的低18%,冷藏3個月後則降低35%。
⒋ 接種量
zhibeibaozishidejiezhongliangyaoshizhong,jiezhongliangguodahuoguoxiaojunduibaozizhiliangchanshengyingxiang。yinweijiezhongliangdedaxiaoyingxiangdaozaiyidingliangpeiyangjizhongbaozidegetishuliangdeduoshao,jineryingxiangdaojuntideshenglizhuangtai。fanjiezhonghoujunluojunyunfenbuzhenggexiemian,yinyuekefenjunluozheweizhengchangjiezhong。jiezhongliangguoxiaozexiemianshangchangchudejunluoxishu,jiezhongliangguodazexiemianshangjunluomijiyipian。yibanchuandaiyongdexiemianbaoziyaoqiujunluofenbujiaoxi,shiyutiaoxuandangejunluojinxingchuandaipeiyang。jiezhongyaopinghuojinguandexiemianbaozi,yaoqiujunluomidushizhonghuoshaomi,baozishudadaoyaoqiubiaozhun。yibanyizhigaoduwei20 cm、直徑為3 cm的試管斜麵,絲狀菌孢子數要求達到107以上。
jieruzhongziguandebaozijiezhongliangduifajiaoshengchanyeyouyingxiang。liru,qingmeisuchanshengjunzhiyideqiuzhuangjundebaozishuliangduiqingmeisufajiaochanliangyingxiangjida,ruobaozishuliangguoshao,zejinguanhouchangchudeqiuzhuangtiguoda,yingxiangtongqixiaoguo;若孢子數量過多,則進罐後不能很好地維持球狀體。
除了以上幾個因素需加以控製之外,要獲得高質量的孢子,還需要對菌種質量加以控製。用各種方法保存的菌種每過1年都應進行1次自然分離,從中選出形態、生產性能好的單菌落接種孢子培養基。製備好的斜麵孢子,要經過搖瓶發酵試驗,合格後才能用於發酵生產。
㈡ 影響種子質量的因素及其控製
種子質量主要受孢子質量、培養基、培養條件、種齡和接種量等因素的影響。搖瓶種子的質量主要以外觀顏色、效價、菌絲濃度或黏度以及糖氮代謝、pH值變化等為指標,符合要求方可進罐。
種子的質量是發酵能否正常進行的重要因素之一。種子製備不僅是要提供一定數量的菌體,更為重要的是要為發酵生產提供適合發酵、具有一定生理狀態的菌體。種子質量的控製,將以此為出發點。
⒈ 培養基
種(zhong)子(zi)培(pei)養(yang)基(ji)的(de)原(yuan)材(cai)料(liao)質(zhi)量(liang)的(de)控(kong)製(zhi)類(lei)似(si)於(yu)孢(bao)子(zi)培(pei)養(yang)基(ji)原(yuan)材(cai)料(liao)質(zhi)量(liang)的(de)控(kong)製(zhi)。種(zhong)子(zi)培(pei)養(yang)基(ji)的(de)營(ying)養(yang)成(cheng)分(fen)應(ying)適(shi)合(he)種(zhong)子(zi)培(pei)養(yang)的(de)需(xu)要(yao),一(yi)般(ban)選(xuan)擇(ze)一(yi)些(xie)有(you)利(li)於(yu)孢(bao)子(zi)發(fa)芽(ya)和(he)菌(jun)絲(si)生(sheng)長(chang)的(de)培(pei)養(yang)基(ji),在(zai)營(ying)養(yang)上(shang)易(yi)於(yu)被(bei)菌(jun)體(ti)直(zhi)接(jie)吸(xi)收(shou)和(he)利(li)用(yong),營(ying)養(yang)成(cheng)分(fen)要(yao)適(shi)當(dang)地(di)豐(feng)富(fu)和(he)完(wan)全(quan),氮(dan)源(yuan)和(he)維(wei)生(sheng)素(su)含(han)量(liang)較(jiao)高(gao),這(zhe)樣(yang)可(ke)以(yi)使(shi)菌(jun)絲(si)粗(cu)壯(zhuang)並(bing)具(ju)有(you)較(jiao)強(qiang)的(de)活(huo)力(li)。另(ling)一(yi)方(fang)麵(mian),培(pei)養(yang)基(ji)的(de)營(ying)養(yang)成(cheng)分(fen)要(yao)盡(jin)可(ke)能(neng)地(di)和(he)發(fa)酵(jiao)培(pei)養(yang)基(ji)接(jie)近(jin),以(yi)適(shi)合(he)發(fa)酵(jiao)的(de)需(xu)要(yao),這(zhe)樣(yang)的(de)種(zhong)子(zi)一(yi)旦(dan)移(yi)入(ru)發(fa)酵(jiao)罐(guan)後(hou)也(ye)能(neng)比(bi)較(jiao)容(rong)易(yi)適(shi)應(ying)發(fa)酵(jiao)罐(guan)的(de)培(pei)養(yang)條(tiao)件(jian)。發(fa)酵(jiao)的(de)目(mu)的(de)是(shi)為(wei)了(le)獲(huo)得(de)盡(jin)可(ke)能(neng)多(duo)的(de)發(fa)酵(jiao)產(chan)物(wu),其(qi)培(pei)養(yang)基(ji)一(yi)般(ban)比(bi)較(jiao)濃(nong),而(er)種(zhong)子(zi)培(pei)養(yang)基(ji)以(yi)略(lve)稀(xi)薄(bo)為(wei)宜(yi)。種(zhong)子(zi)培(pei)養(yang)基(ji)的(de)pH值要比較穩定,以適合菌的生長和發育。pH值的變化會引起各種酶活力的改變,對菌絲形態和代謝途徑影響很大。例如,種子培養基的pH值控製對四環素發酵有顯著影響。
⒉ 培養條件
種zhong子zi培pei養yang應ying選xuan擇ze最zui適shi溫wen度du,前qian麵mian已yi有you敘xu述shu。培pei養yang過guo程cheng中zhong通tong氣qi攪jiao拌ban的de控kong製zhi很hen重zhong要yao,各ge級ji種zhong子zi罐guan或huo者zhe同tong級ji種zhong子zi罐guan的de各ge個ge不bu同tong時shi期qi的de需xu氧yang量liang不bu同tong,應ying區qu別bie控kong製zhi,一yi般ban前qian期qi需xu氧yang量liang較jiao少shao,後hou期qi需xu氧yang量liang較jiao多duo,應ying適shi當dang增zeng大da供gong氧yang量liang。在zai青qing黴mei素su生sheng產chan的de種zhong子zi製zhi備bei過guo程cheng中zhong,充chong足zu的de通tong氣qi量liang可ke以yi提ti高gao種zhong子zi質zhi量liang。例li如ru,將jiang通tong氣qi充chong足zu和he通tong氣qi不bu足zu兩liang種zhong情qing況kuang下xia得de到dao的de種zhong子zi都dou接jie入ru發fa酵jiao罐guan內nei,它ta們men的de發fa酵jiao單dan位wei可ke相xiang差cha1倍。但是,在土黴素發酵生產中,一級種子罐的通氣量小一些卻對發酵有利。通氣攪拌不足可引起菌絲結團、junsizhanbidengyichangxianxiang。shengchanguochengzhong,youshizhongzipeiyanghuichanshengdaliangpaomoeryingxiangzhengchangdetongqijiaoban,cishiyingyangekongzhi,shenzhikekaolvgaibianpeiyangjipeifang,yijianshaofapao。
對(dui)青(qing)黴(mei)素(su)生(sheng)產(chan)的(de)小(xiao)罐(guan)種(zhong)子(zi),可(ke)采(cai)用(yong)補(bu)料(liao)工(gong)藝(yi)來(lai)提(ti)高(gao)種(zhong)子(zi)質(zhi)量(liang),即(ji)在(zai)種(zhong)子(zi)罐(guan)培(pei)養(yang)一(yi)定(ding)時(shi)間(jian)後(hou),補(bu)入(ru)一(yi)定(ding)量(liang)的(de)種(zhong)子(zi)培(pei)養(yang)基(ji),結(jie)果(guo)種(zhong)子(zi)罐(guan)放(fang)罐(guan)體(ti)積(ji)增(zeng)加(jia),種(zhong)子(zi)質(zhi)量(liang)也(ye)有(you)所(suo)提(ti)高(gao),菌(jun)絲(si)團(tuan)明(ming)顯(xian)減(jian)少(shao),菌(jun)絲(si)內(nei)積(ji)蓄(xu)物(wu)增(zeng)多(duo),菌(jun)絲(si)粗(cu)壯(zhuang),發(fa)酵(jiao)單(dan)位(wei)增(zeng)高(gao)。
